Effects of β-catenin on Regnlating Hair Follicle Cell′s Proliferation

Abstract

Objective To study the mechanism of β-catenin on the regulation of proliferation and differentiation of hair follicle cells. Methods Aminoterminal truncated β-catenin gene was transfected into hair follicle cells. The expression of β-catenin was detected by immunocytochemistry to identify the success of transfection. Proliferation of transfected cells was measured by clone forming efficiency. The expression of keratin 19 of transfected cells was detected by immunocytochemistry. The expression of the target gene c-myc was observed by Western blotting and in situ hybridization. Results The expression of β-catenin in the nuclear after transfection increased indicating the success of aminoterminal truncated β-catenin gene transfection. Clone forming efficiency increased significantly in transfected cells, however, there was no significant increase of large clone. The expression of K19 increased slightly. Both of the expression of c-Myc protein and mRNA increased significantly after transfection, suggesting that the increase of β-catenin promoted the transcription and translation of c-myc gene.
